Clinical and Interpretive
Initially, the DAT will be performed using a poly-specific regent capable of detecting both IgG and C3D. If the results are positive, the DAT will be tested with a reagent containing IgG only to appropriately characterize the immune process. This is a test to demonstrate the presence of immunoglobulin on the surface of the red cells. It is used in the investigation of hemolytic anemia and in conjunction with other testing and clinical data can characterize hemolysis as immune-mediated.
IgG antibody may be present on patients or transfused RBCs and may cause hemolysis. The antibodies may be directed against self-antigens (autoimmune hemolysis), maternal antigens (hemolytic disease of the newborn), donor antigens (alloimmune transfusion reaction), and drugs.
This assay tests for the presence of in vivo coating of patient RBCs by IgG.
